Influencia de la actinomicina D sobre el flujo sanguíneo cerebral en un modelo de isquemia experimental

Existen estudios experimentales sobre el mejoramiento del vasoespasmo que sigue a la hemorragia subaracnoidea por la Actinomicina D, pero no existe ningún estudio acerca de su efecto en la isquemia cerebral.

Hemos realizado el presente estudio experimental en 18 ratas; produciéndoles isquemia cerebral parcial (mediante ligadura permanente de la arteria carótida común derecha, y clipaje transitorio de la carótida común izquierda), que fueron tratadas con Actinomicina D (vía intraperitoneal) por 5 días. Se administró dicha sustancia a las 6 y 12 horas postisquemia, en los grupos A (6 ratas) y B (6 ratas) respectivamente. El grupo C (control) constituído también por 6 ratas, no recibió tratamiento alguno. El análisis estadístico antes y después del tratamiento se realizó observando las modificaciones del flujo sanguíneo cerebral utilizando HMPAO marcado con Tc-99m (marcador radioactivo).

Se encontró ausencia de modificación del flujo sanguíneo cerebral, y por lo tanto ausencia de mejoría de la isquemia cerebral.

Se concluye que la Actinomicina D no modifica el flujo sanguíneo ni mejora la isquemia cerebral experimental en ratas.

Though there are some studies reporting a favourable effect of Actinomycin D posthemorrhagic cerebral vasospasm, the effect of this drug on brain ischemia has not been analyzed.

We have performed an experimental study in 18 rats. Ischemia was produced by permanent and transitory ligature of the right and left common carotid artery respectively.

The animals were divided into three groups. Actinomycin was injected intraperitoneally 6 and 12 hours after ischemia in groups A and B. Animals in group C did not receive any treatment.

Cerebral blood flow (CBF) was measured using HMPAO labeled with Tc 99m.

There was no modification of CBF. The conclusion is that Actinomycin D does not improve cerebral ischemia in this experimental model.
