Estudio morfométrico de los núcleos supraóptico y paraventricular tras estímulos dolorosos y administración de morfina intracerebroventricular

Resumen

El objetivo de este trabajo es estudiar los cambios de la superficie neuronal de los núcleos supraóptico (NSO) y paraventricular (NPV) en la rata tras aplicar estímulos dolorosos y tratamiento con morfina intracerebroventricular (ICV). Para ello se estudiaron 7 grupos de 3 animales cada uno divididos de la siguiente forma: grupo control, grupo testigo (implante de cánula ICV), grupo dolor (administración subcutánea de 10mcl de formaldehido), grupo morfina (administración ICV de 30mcg de morfina), grupo dolor-naloxona (formaldehido subcutáneo más 0.1mg de naloxona i.m.) y finalmente, grupo dolor-naloxona-morfina. La técnica histológica empleada fue la de hematoxilina crómica. El estudio morfométrico se realizó sobre 100 células de cada grupo escogidas aleatoriamente y su superficie fue medida mediante un procesador digital de imágenes. Los resultados obtenidos indican que 1) el dolor produce un aumento significativo de la superficie neuronal de los NSO y NPV, siendo en orden decreciente según los grupos dolor, dolor-naloxona, dolor-naloxona-morfina y 2) la morfina provoca una disminución significativa en el área neuronal del NSO.

Summary

The aim of the present work was to carry out a morphometric study of the effects of painful stimulation and morphine treatment on the hypothalamic supraoptic and paraventricular nuclei in the rato Neurons were stained using chromic-haematoxilin stain.

The results obtained have been evaluated by using statistical methods. Our conclusions were: 1) Met-enkephaline is synthesized in aH the magnoceHularis neurons; 2) Responses observed in hypothalamic magnoceHularis neurons in response to painful stim.ulation, morphine treatment or naloxone injection are quantitative and stimulus-dependent; 3) The response is more intense in neurons from the supraoptic nucleus than in neurons from the paraventricular nudeus.
